OPPORTUNITIES ARISING FROM THE SEQUENCING THE GENOME OF THE SIREX PARASITIC NEMATODE, DELADENUS SIRICIDICOLA Chongxing Zhang, X. Osmond Mlonyeni, Alisa Postma, Brenda D. Wingfield, Michael J. Wingfield, Bernard Slippers* Department of Genetics, Forestry and Agricultural Biotechnology Institute, University of Pretoria *bernard.slippers @fabi.up.ac.za INTRODUCTION Across the Southern Hemisphere Sirex noctilio is the most invasive pest of pine trees causing devastating economic losses to the forestry industry The entomoparasitic nematode, Deladenus siricidicola, a cornerstone of the biological control program against S.noctilio has had varying levels of parasitism success (Hurley et al., 2007) Microsatellite analysis revealed D. siricidicola populations within the Southern Hemisphere to be highly inbred (Mlonyeni et al., 2011) To date, genomic studies of seven nematodes species have provided an opportunity to investigate nematode genomic diversity, nature of mutualistic and parasitic lifestyles and process involved in ecological adaptation To explore and understand the underlying mechanism of parasitism and evolution of virulence, we are sequencing the genome of D. siricidicola OBJECTIVES 1. 2. 3. 4. Characterize the phenotypic and molecular diversity of Deladenus siricidicola from around the world and assess its potential of influencing ecological interactions and control efficiency Characterize the genetic basis for variation in virulence in D. siricidicola Characterize the genetics of virulence evolution in D. siricidicola Propose tools to more effectively monitor Sirex noctilio, as well as to maximize effects of evolution on virulence for the control of S. noctilio MATERIALS AND METHODS DNA and RNA extraction The Deladenus siricidicola Kamona strain was used for the sequencing project. Nematodes were harvested from cultures grown in 500ml Erlenmeyer flask containing wheat, rice and Amylostereum areolatum. Genomic DNA was extracted using the phenolchloroform method. Total RNA was extracted using RNeasy® Plant Mini Kit. Sequencing 454 Life Science/Roche GS FLX sequencing system was used to sequence the D. siricidicola genome at Inqaba Biotec? (Pretoria, South Africa). Both shortgun and paired-end libraries were used for this sequencing. In addition, SoLiD sequencing platform at SeqOmics Ltd (Hungary) was used to sequence the genome and transcriptome of D. siricidicola. For this genome sequencing, 50+50 mate pairs sequences were used. Table 1.
